    How to Use a Peptide Reconstitution Calculator

    A peptide reconstitution calculator converts three user-supplied values — vial amount, added liquid volume and target research amount — into a concentration, a dose volume in millilitres and the equivalent U-100 syringe units.

    The three values you supply

    Every reconstitution calculation starts with numbers that come from your own vial label and your own protocol notes. The calculator does not know or assume any of them.

    • Vial amount — the labelled quantity of lyophilised peptide, usually in milligrams (mg).
    • Diluent volume — how much bacteriostatic water or other liquid you add, in millilitres (mL).
    • Target research amount — the quantity you intend to withdraw per draw, in mg or mcg.

    The arithmetic

    The calculation is pure division. Concentration is the vial amount spread across the liquid you added; dose volume is the target amount divided by that concentration.

    Concentration (mg/mL) = vial amount (mg) ÷ diluent volume (mL)

    Dose volume (mL) = target amount (mg) ÷ concentration (mg/mL)

    U-100 syringe units = dose volume (mL) × 100

    A worked example

    A 10 mg vial reconstituted with 2 mL of bacteriostatic water gives 10 ÷ 2 = 5 mg/mL. A target amount of 0.5 mg then needs 0.5 ÷ 5 = 0.1 mL, which is 0.1 × 100 = 10 units on a U-100 insulin syringe.

    Note what did not change: the vial still contains 10 mg of peptide. Adding more liquid lowers the concentration and increases the volume you draw — it never changes the total amount in the vial.

    Verifying your own inputs

    The output is only as accurate as the values entered. Confirm the labelled vial amount, confirm the units printed on your syringe barrel (U-100 syringes are marked 0–100 units for 0–1 mL), and re-read the diluent volume you actually added rather than the volume you intended to add.

    Frequently asked questions

    Does adding more bacteriostatic water change the total peptide amount?

    No. The vial contains a fixed mass of peptide. Adding more liquid lowers the concentration in mg/mL and increases the volume required for the same amount; the total in the vial is unchanged.

    Why do two calculators sometimes show different syringe units?

    Almost always because of a different assumed syringe scale (U-100 vs U-40), a different unit for the target amount (mg vs mcg), or rounding at a different decimal place. Check all three before assuming one calculator is wrong.
