    Tesofensine

Research Use OnlyInvestigational New Drug (US)

    Tesofensine

    Description

A serotonin-norepinephrine-dopamine reuptake inhibitor originally studied as an anti-obesity agent. Promotes weight loss through appetite suppression.

    Dose:0.25-1 mg
    Frequency:Daily
    Administration Method: Oral
    CategoryWeight Loss
    Half-Life~220 hours

    Overview

Tesofensine was originally studied for neurodegenerative disease and was found to induce weight loss by reducing hunger and increasing satiety, shifting the focus toward obesity treatment.

    Mechanism of Action

Tesofensine is a triple monoamine reuptake inhibitor (serotonin, noradrenaline, dopamine - SNDRI) that modulates appetite, energy expenditure, and motivation.

    Benefits

    • •Weight loss
    • •Appetite suppression
    • •Metabolic enhancement
    • •Energy increase
    • •Fat burning

    Dosing Notes

Most weight-loss trials evaluated oral doses of 0.25, 0.5, and 1 mg/day. 0.5 mg/day showed the most balanced efficacy/tolerability; 1 mg/day produced more weight loss but more side effects.

    Reported Side Effects

    • •dry mouth
    • •constipation
    • •insomnia
    • •increased heart rate
    • •mild changes in blood pressure

    Safety Notes

Side effects increase with dose. Because tesofensine has a very long half-life, adverse effects may persist if the dose is too high.
